Although most luminescent reporter methods are destructive to the cell samples, luminescence from living cells can be measured by adding substrates to the growth medium. As previously reported, two luminescent signals were measured simultaneously in living cells by using Chroma-Luc™ luciferases capable of emitting green and red light. We now report an extension of the color separation between the luciferases by using the blue emission of Renilla luciferase (480nm) with the red emission of the Chroma-Luc™ luciferase (611nm). The combined light emission can be achieved using EnduRen™ Live Cell Substate and beetle luciferin.
